I have horses E, C, and D, but am curious if the big “?” horse is possible, or if it will be blocked by EV’s inbreeding block? 
Has anyone tried this or know if it could work? i can test it myself in a few weeks once I have horse F, but i’m impatient and eager to know now!

It should work, the block applies up to the grandparent side.
